At the present day, B chromosomes still remain one of the most mysterious karyotypic elements of eukaryotes. For many years studies of B chromosomes had been limited to analysis of their morphology, frequencies in natural populations, effects on host fitness, behavior in mitosis and meiosis. Development of new techniques of molecular cytogenetic analysis allowed starting studies and comparison of B chromosomes at the level of their DNA composition. In this study morphological and molecular organization of B chromosomes of the Korean field mouse Apodemus peninsulae and two species of grasshopper (Podisma sapporensis and Podisma kanoi) was analysed with Giemsa, DAPI, Ag-NOR staining, and chromosome painting with the whole and partial chromosome probes generated by microdissection and DOP-PCR. We analyzed DNA composition of the B chromosomes belonging to different morphotypes, and studied distribution of repeats homologous to B chromosomal DNA on A chromosomes. Based on obtained data, the role of B chromosomes in karyotypic evolution and evolutionary pathways of their origin and further development are discussed.
