A brief history of the study of the fauna and taxonomy of Russian margaritiferids is presented, as well as a critical assessment of the use of various conchological characters. Based on the analysis of shell morphology of type specimens and literature data on phylogeny, the taxonomic position, synonymy and distributional areas of pearl mussels are clarified. The list of Margaritiferidae species included in the newly published Red Data Book of the Russian Federation has been corrected as follows: Margaritifera margaritifera (L., 1758) (syn.: Unio elongata Lamark, 1819; Margaritana borealis Westerlund, 1871); Margaritifera dahurica (Middendorff, 1850) (syn.: Dahurinaia sujfunensis Moskvicheva, 1973; D. tiunovae Bogatov et Zatravkin, 1988; D. komarovi Bogatov, Prozorova et Starobogatov, 2003; D. ussuriensis Bogatov, Prozorova et Starobogatov, 2003; D. prozorovae Bogatov et Starobogatov, 2003 in Bogatov, Prozorova et Starobogatov, 2003; D. transbaicalica Klishko, 2008); Margaritifera laevis (Haas, 1910) (syn.: Margaritana sachalinensis Zhadin, 1938); Margaritifera middendorffi (Rosen, 1926) (syn.: Dahurinaia shigini Zatravkin et Bogatov, 1987 syn. nov.; Kurilinaia kamchatica Bogatov, Prozorova et Starobogatov, 2003; K. zatravkini Bogatov, Prozorova et Starobogatov, 2003); Margaritifera kurilensis (Zatravkin et Starobogatov, 1984) (syn.: M. togakushiensis Kondo et Kobayashi, 2005 syn. nov.). It is shown that the synonymy between M. togakushiensis and M. middendorffi is erroneous, because their type specimens have different structure of cardinal teeth. The latter species may be a junior synonym for M. laevis.
